Bremelanotide, catalogued in the research literature as PT-141, is a synthetic cyclic heptapeptide of the melanocortin class, structurally derived from the His-D-Phe-Arg-Trp core pharmacophore of the melanocyte-stimulating hormone family. Its covalent structure is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH: an N-acetylated norleucine residue precedes a macrolactam ring closed between the aspartate side-chain carboxyl and the lysine side-chain amine, and the peptide terminates in a free carboxylic acid rather than a carboxamide, which distinguishes it from the closely related cyclic lactam melanotan II. The molecular formula is C50H68N14O10 and the average molecular weight of the free base is 1025.2 g/mol (PubChem CID 9941379; CAS 189691-06-3). The free base is a white to off-white lyophilized solid; the arginine, histidine and free-acid functionalities give a highly polar, water-soluble peptide, and the substance is frequently catalogued as an acetate salt.

The compound is characterized in the literature as a non-selective agonist across the melanocortin receptor subtypes, and it recurs as a comparator ligand in structure-activity studies of macrocyclic melanocortin scaffolds assayed in receptor-transfected cell lines (Yuan and Tao, 2022; Merlino et al., 2026). Its degradation chemistry, including epimerization pathways, has been characterized analytically by LC-HRMS/MS with accompanying computational modelling (Yuvaraaj and Sharma, 2026). In cell-culture systems, melanocortin-receptor-dependent changes in survivin expression and cell viability have been examined in glioblastoma lines (Suzuki et al., 2024). Non-clinical central nervous system pharmacology has been reviewed for rodent models (Pfaus et al., 2007), and behavioural and neural-activation endpoints have been examined in Syrian hamster models (Borland et al., 2025).
